Today we have a special one! In this episode, we have Vitalik Buterin as a guest, interviewed by Vincent Weisser. They discuss the intersection of science and cryptocurrency, focusing on the concept of "decentralized science." 
Vitalik explains how decentralized technologies, such as cryptocurrency, can improve science and the institutions of science by allowing for more diverse funding and collaboration among people. Welcome to Episode Nr. 4 of the DeSci Podcast - Today we're exploring the decentralized science space together with Jack Scannell! Jack coined the term Eroom's Law to describe the long term exponential decline in pharmaceutical R&D efficiency. It contrasts with Moore’s Law which describes the exponential increase in computer chip efficiency. “Eroom” is “Moore” spelled backwards.

In this podcast, Jack talks about how we can do better drug discovery, specifically, the tools and frameworks we can use to choose the right drug discovery model and feed that into investment decisions. His new paper in Nature Reviews Drug Discovery addresses exactly that: https://rdcu.be/cWTtO

Jack argues that quality beats quantity when it comes to assays (though the ideal is quality AND quantity). So it's better to test a small number of therapeutic candidates in a really good model than it is to test a very large number of candidates selected from the same universe using a model with low validity. A clear example comes from antimicrobial drug discovery. Screening a few hundred compounds in mouse sepsis models in the 1930s appears to have been more productive than testing over ten million compounds against isolated bacterial gene products in the 1990s and early 2000s.

The main bottleneck in drug R&D has shifted from chemistry to the “predictive validity” of the tools that tell you whether or not that chemistry is useful. We can make plenty of compounds and identify lots of targets, but we lack the tools to tell us whether those compounds and targets are meaningful in human disease. This is addressable. There are frameworks to evaluate the likely quality of decision tools (e.g., screening and disease models). These should be more rigorously developed and applied. A great deal of model choice simply follows from tradition or availability. We should start by defining what a good model should look like (a “Target Model Profile”) and then evaluate models against that profile before deciding whether or not to believe their results. 

Jack on what interested him about VitaDAO and Molecule: You create the most value not by making current markets a little bit more efficient, but by making markets exist where none existed before. Outside of a few well-ventured universities, academics are under-ventured and also the people at those universities don't have the right tools to present their material in such a way that it could be ventured so it struck him that providing a low transaction cost, standardized platform to make an “eBay for IP” might be an extremely valuable thing to do because you could create markets where none existed before.

In this episode, we discuss the problem of declining R&D efficiency with Dr. Jack Scannell, a scientist and entrepreneur who is well known for coining the term “Eroom’s law,” which highlights the decline in pharmaceutical research and development (R&D) productivity over several decades.

In this episode, Jack sheds light on the root causes of this decline, beginning with a critique of the poor quality of diagnosis surrounding the R&D productivity problem.
He also talks about the challenges in accurately measuring progress in drug research, highlighting the "measurement problem" in the drug industry. Jack suggests adopting a model akin to the "Stern Review” to improve the diagnosis of these challenges.
